Abstract: PURPOSE: This technical note provides protocols for maintenance of cell cultures and for the conduct of a biomarker-based screening assay for dioxin toxic equivalents (TCDD TEQs) in sediments and other environmental samples using the H4IIE rat hepatoma cell line as performed at the U.S. Army Engineer Waterways Experiment Station (WES). The purpose of these protocols is to provide analysts who may wish to gain the capability for performing the assay with a detailed “blueprint” of how the assay is conducted and of what is necessary in terms of technique, materials, instrumentation, and time.
